Vitamin D receptor antagonism on mRNA expression of reproductive hormones at hypothalamic and pituitary level of adult male rats

Abstract:
Objective: To investigate mRNA expression of GnRH in hypothalamic fragments and gonadotropins in pituitary glands of adult male Sprague Dawley rats after vitamin D receptor antagonism by ZK159222. Methodology: This randomized prospective controlled trial was carried out in Lab of Reproductive Neuroendocrinology, Department of Zoology, Quaid-i-Azam University, Islamabad in collaboration with Department of Physiology Islamic International Medical College, Riphah International University Islamabad, Pakistan from January December 2018. Twenty rats were procured from National Institute of Health, Islamabad. Hypothalami and pituitaries of these rats were collected and were equally divided into control (C) and experimental (T) groups. Experimental group tissues were exposed to 200nM concentration of vitamin D receptor antagonist (ZK159222). After incubation, tissues of both groups were flash frozen and stored at -80ºC for expression analyses. Student’s t- test was used for analyses. Results: Real time qPCR showed up regulation of mRNA of gonadotropin releasing hormone (GnRH) and down regulation of luteinizing hormone (LHß) mRNA after VDR antagonist treatment. Conclusion: The preliminary data suggests that vitamin D has modulatory effect on expression mRNA of GnRH in hypothalamic fragments and FSHß and LHß in pituitary gonadotrophs.
