Molecular phylogeny and morphological characterization revealed a new species in Lepiota from Margalla Hills, Pakistan

Abstract:
Lepiota is a large and diverse genus of white-spored Agaricales and litterinhabiting fungi which commonly occur in the tropics. It is represented by more than 500 species distributed all over the world. Members of this genus are characterized by fusiform-amygdaliform basidiospores with convex abaxial and convex adaxial sides, or with a straight abaxial side, in combination with a pileus covering made up of long cylindrical elements with or without short clavate elements at the base and presence of clamp connections. Pileus covering and basidiospores shape are the key characteristics that divide this genus into several sections. From Pakistan, more than 30 species have been reported. However, there are still a lot of species that need to be identified and described. Specimens were collected from the Margalla hills, Pakistan which were characterized morpho-anatomically and using DNA sequences of internal transcribed spacer regions (ITS) of nuclear ribosomal RNA gene. During current investigation, this species found closely related to species in Lepiota but its comparative study based on morphology and phylogeny revealed that it is distinct from already known taxa in the genus and represent a new species from Pakistan.
