An overview of strategies for synthetic modifications of aminoglycosides.

Abstract:
Aminoglycosides, a family of structurally related broad-spectrum bactericidal compounds, have been used extensively for the treatment of aerobic gram-negative bacterial infections because of their rapid anti-bacterial activity, chemical stability and ability to work in combination with other drugs. However, toxic side-effects and growing bacterial resistance have narrowed the significance of aminoglycosides as antibiotics. Due to these limitations, extensive research on aminoglycosides has resulted in the development of a wide range of synthetic strategies to improve the overall characteristics of aminoglycosides. Herein, an overview of the various approaches reported in the literature to enhance binding affinity and to overcome resistance of aminoglycosides is provided.
