Pharmacological profiling and phytochemical analysis of fractionated extracts of Euphorbia royleana

Abstract:
Background: Euphorbia royleanais a member of family Euphorbiaceae, with great therapeutic potential. Present study is aimed to validate its traditional uses. Euphorbia royleanacrude and fractionated extracts were subjected to cytotoxic, antioxidants, antibacterial and phytochemical analysis. Extraction was carried out using cold maceration technique. Cytotoxic screening of various extracts of selected plant was performed by Brine shrimp lethality (BSL) assay. Phytochemicals in fractionated extracts were identified and quantified using standard protocols. Free radical scavenging activity was evaluated using 2, 2diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6sulfonic acid (ABTS), Total antioxidant assay (TAC) and Ferric Reducing Antioxidant Power (FRAP) assays. Elemental analysis of plant was performed by employing atomic absorption spectroscopy. Antibacterial activity of plant extracts were tested on selected bacterial strains (E.coli, Klebsiella, Xanthomonas and Staphylococcus aureus) with agar well diffusion assay. Among all fractionated extracts of tested plant, crude and hexane extract found to possess the highest mortality rate (80 and 70 % respectively at 400µg/ml). The phytochemical analysis confirmed the presence of phenols, glycol cyanide, tannins, saponins, alkaloids and flavonoids were found in both crude and fractionated extracts (1.886, 6.001, 2.231, 0.884, 0.092 and 0.551 µg/mg respectively). Plant extracts also showed scavenging activity range from 23-61% at 25-400 µg/ml. E. royleanais a source of variety of minerals, namely K +, Na+, Fe+2, Co+3, Mn+2, Cu+3, Cr+3, and Cd+2. Plant extracts exhibited antibacterial activity against selected bacterial strains. Results were evaluated statistically.
