Role of Transcription Regulators for Differentiation of Mesenchymal Stem Cells into Oligodendrocyte-Like Cells

Abstract:
Myelin sheath is involved in providing metabolic support, electrical insulation and transmission of action potential through axons. Injury or inflammation to the CNS may lead to the degeneration of this protective covering resulting in the loss of oligodendrocytes, which are the type of glial cells involved in the formation of myelin. For this purpose, stem cell replacement therapy is an alternative and a progressive approach in the field of regenerative medicines. Stem cells have the potential to differentiate into multiple cell lineages by secreting trophic and immunomodulatory factors.Therefore, in the proposed research, our aim was to generate oligodendrocyte like cell lineage to assess their capability into myelinating oligodendrocytes by inducing transcription regulators MYT1L and OLIG2 into human umbilical cord stem cells with the help of transfection reagent. Our results indicated protein expression of oligodendrocytes by immunocytochemistry which was confirmed through gene expression analysis by qPCR. Our study concluded that these transcription regulators possess therapeutic effect in treating demyelinating diseases in CNS. Through our study, we found an alternative approach to overcome the consequences produced by demyelination diseases and were successful in designing effective cell-based strategy for its treatment.
