A study in callogenesis of date palm seedling tissue.

Abstract:
Shoot and root explants of date palm seedlings were cultured on various media. Shoot apical segments formed callus on MS containing 0.1 mg/1 2, 4-D and 0.1 mg/1 K with 1000 mg/1 CH. Root explants formed callus on lateral root tips on AM medium containing 2,4-D,BAP, K, AS, NAA and CH at 6. 0,0. 1,0. 3,4. 0,1.0 and. 1000 mg/1 respectively. It was observed that an alternate temperature of 28°C (day) and 30°C (night) with 74. 3% RH was required for callogenesis in shoot explant, while 34. 3 and 27.5°C. with 48% RH was required for callogenesis in root explants.
