Abstract:
Geminiviridaea is one of the most important family of plant-infecting viruses that has been classified into fourteen recognized genera. Amongst these, Begomovirus is the largest and economically most important genus. Viruses of the genus are further categorized into mono partite (single genomic component or DNA-A) and bipartite (two genomic components called DNA-A and DNA-B)begomoviruses. These viruses infect a wide range of plant species in tropical and sub-tropical regions of the world. Different plants such as Amaranthusretroflexus, Cannabis sativa, Capsicum annum, Glycine max, Mentha longifolia, Parthenium hysterophorus that showed symptoms indicative of begomo virus infection were sampled in several districts of Khyber Pakhtunkhwa.Begomo viruses, master viruses and their associated alpha satellite and/orbeta satellite present in these samples were amplified by RCA and PCR, cloned, and sequenced. The phylogenetic and biogeographic analyses of viruscomponents were performed by various bioinformatics tools. Complete genome sequences of begomoviruses and satellites obtained in this study showed the highest sequence identity with chili leaf curl virus, tomato leaf curl virus, tomatoleaf curl New Delhi virus, chili leaf curl beta satellite, parthenium leaf curl alpha satellites and papaya leaf curl beta satellite. Tomato leaf curl virus and Tomato leaf curl New Delhi virus were the prominent viruses ,whereas, Papaya leaf curl beta satellite and Parthenium leaf curlalpha satellite were the prominent beta satellites and alpha satellite, respectively in the region. The existence of these viruses/ satellites in numerous diverse important plant hosts might be a sign that future epidemics are possible in thearea and is an alarming signal for future food security in the region. The study also provides an understanding of the origin and evolution of both mono partite and bipartite begomo viruses and highlights the need for further research on developing management strategies.
