Immobilization of Fungal Glucoamylase Using Calcium Alginate Beads

Abstract:
Enzymes have remarkable advantages over chemical catalysts but their use on industrial scale is vulnerable due to decrease in their stability, efficiency and due to harsh environmental conditions. Current study is a challenge to increase the stability and efficiency of gluco amylase by its immobilization through entrapment method. For this purpose, gluco amylase was produced from previously isolated specie of Aspergillus niger and immobilized on sodium alginate by an entrapment method. Various parameters were used namely sodium alginate concentrations, calcium chloride concentrations, beads surface area, optimization of pH and temperature, reaction time, substrate concentration and reusability of enzyme were optimized and associated with the free enzyme. The maximum immobilization yield was obtained at 4% concentration of sodium alginate, 0.3M calcium chloride concentration and 4 cm beads diameter. The optimum pH for immobilized and free enzymes were 4 and 5correspondingly, temperature remained 50 °C and 60 °C and reaction time was10 minutes. Thus, it can be expected from current findings that sodium alginate immobilized gluco amylase can be used to produce commercially important products and efficiently for hydrolysis of starch.
