Taxonomic and phytochemical analysis of Gemini virus infected and healthy Duranta erecta (L.)

Abstract
Duranta eracta L. is widely grown as an ornamental and medicinal plant belongs to the family Verbenaceae. In the current study, various surveys were conducted for the observation and collection of Gemini viruses infected and non-infected plants of D. erecta L. from different areas of Lahore. Collected samples were compared for taxonomic, antioxidant, phytochemicals, molecular and antimicrobial potential. Morphological analysis based on characters like shape, size, veins thickening, colour of leaves indicated that infected leaves exhibited curling, cupping, veins thickening and stunted growth. In anatomical analysis, size of epidermal cells, presence and absence of stomata, guard cells, subsidiary cells and trichomes were observed on abaxial and adaxial surfaces. Healthy leaves showed more length and width of dermal cells as compared to infected leaves. Phytochemical analysis indicated Cardiac glycosides, tannins, terpenoids and saponins were present in both healthy and infected samples but anthraquinones and phlobatannins were absent in both samples. Antioxidant potential was measured by 2, 2 diphenyl-1 picrylhydrazyl by spectrophotometer at 517nm which was highest in methanol, moderate in ethanol and chloroform, minimum in n-hexane extracts. Geminiviruses infection was confirmed by using PCR technique, in which universal diagnostic primers CLCV1 and CLCV2 were used. Antibacterial potential was evaluated against Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae which indicated that healthy samplesgave remarkable results (10 mm) against S. aureus while infected samples showed no activity. Observation of antifungal potential against Aspergilus niger, Aspergilus solani, Rhizopus stolonifera give no result in both samples. This is concluded bycurrent studythat healthy samples were found good source of antioxidant and phytochemical potential as compare to unhealthy.
