Abstract:
Vitis jacquemontii is a wild grape, and indigenous people use wild grapes as a substitute for cultivated grapes. Human activities pose a major threat to the plant’s population. Plant tissue culture procedures are recommended for its protection. This study aimed to develop an effective method for inducing callus and shoots and to analyze the chemical constituents in the fruit and leaf extracts. The maximum callus fresh weight (3.12 grams) was achieved on a medium containing naphthalene acetic acid (NAA 1.5 mg/L). The medium supplemented with 1.5 mg/L benzyl adenine (BA) produced a maximum of 8.34 per explant. The medium augmented with NAA had the highest shoot rooting percentage(51.23%) and the largest number of root (5.62). HPLC analysis confirmed eight compounds in the fruit extract (VJFME) and six in the leaf extract (VJLME).Rutine was an abundant compound in the VJLME extract, and quercetin was in the VJFME extract. Quantitative phytochemical analysis revealed higher total tannin (TTC), phenol (TPC), and flavonoids in the VJFME extract than in the VJLME extract. In conclusion, a regenerative system was established to produce materials from V. jacquemontii for bioactive compounds. The phytochemical analysis can enhance its commercial value and serve as a potential antioxidant compound in the future.
