The influence of metal complexing agents on citric acid production by Aspergillus niger.

Abstract:
The present investigation describes the effect of the addition of complexing agents (EDTA and ferrocyanide) on the production of citric acid by Aspergillus niger GCB9 in shake flask. The flasks were rotated at 200 rpm. The incubation temperature was 30+1°C. The mould cultures were screened and the best producer was selected for citric acid fermentation. The parameters, investigated were the concentration of complexing agents on citric acid formation and then time of addition in the fermentation medium. Citric acid production was enhanced by adding EDTA, 0 hour after spore inoculation and ferrocyanide, 24 hour after the inoculation. The amount of citric acid produced was 53.9 g/I and 57.8 g/I, respectively. In the combined effect, addition of both complexing agents was also studied and citric acid production was increased to 64.6 g/I. Effect of the concentration of sugar and different initial pH on citric acid synthesis were also investigated. The optimum level of sugar and pH were 15% and 3.5, respectively. The mycelium in the control media was in form of larger and smaller pellets. It was modified to small rounded pellets after the addition of complexing agents. The supply of oxygen is greatly improved when the growth is in form of small pellets instead of larger pellets.
