Abstract:
Pseudoperithecia grown on seeds were removed and rolled. vigorously on the surface of 3% water agar to clean them of conidia and, other debris prior to crushing. They were then rinsed three times in sterile distilled water, and each pseudoperithecia was transferred to a thin drop of water on a glass microscope slide. There, it was crushed gently with a fine steel needle. Individual asci were separated from the crushed pseudoperithecia and transferred to a thin drop of water on a separate slide using a needle made with the help of a razor blade and dissecting microscope from a longitudinal section of the central rib of a large turkey feather. Ascospores were extruded from the individual asci by pressing and or puncturing them with the fine steel needle. The intermingled spores were separated and each transferred to a petridish of PDA (0.5% agar) using the feather needle developed by Generald Thorne for isolating nematodes.
