A novel design for precise estimation of Michaelis constants.

Abstract:
Human erythrocyte acetylcholinesterase (AChE) activity was assayed in replicate at two concentrations of substrate, one was much lower (S2) and the other much higher (S2) than a prorincial estimate of Km. The Michaelis Menton parameters (Km, and Vm) were computed by a non-linear unweighted regression procedure. This was termed as a "two substrate concentration design" (Ts). A comparison of this design with the more conventional many substrate concentration one (design Ms) showed that the Ts gives more precise and accurate estimates of the parameters than that of Ms. Moreover, the Ts. was found to give more rapid and reproducible answers.
