Impact of Brain Derived Neurotrophic factor gene polymorphism on its peripheral levels in schizophrenic patients

Abstract:
Schizophrenia is a serious, chronic, debilitating neurodevelopmental brain disorder characterized by hallucination and delusions. Prevalence is 1 % worldwide and 1.5 % in Pakistan. It is a complex disorder with diverse etiology and no specific known cause. BDNF is among the candidate genes for schizophrenia and is involved in neurogenesis, synaptogenesis, neuronal plasticity, and modulation of neurotransmitters essential for learning and memory. Aberrant functions of BDNF leads to altered neuronal growth and plasticity. Schizophrenia is a chronic, multigenetic neurodevelopmental brain disorder. BDNF SNP rs12291063 is associated with decreased ventromedial hypothalamic expression of BDNF leading to obesity. As decreased BDNF levels and its polymorphisms are also reported in schizophrenia. It is hypothesized that BDNF polymorphism, rs12291063 might also impact BDNF levels in schizophrenic patients. Therefore, we planned to assess serum levels of BDNF and its polymorphism (rs12291063) in Pakistani schizophrenic patients. Schizophrenia cases (50) were diagnosed using DSM V criteria while controls (50) were individuals without any psychiatric illness. Positive and negative syndrome scale score (PANSS) was used to assess severity of disease. The genomic DNA of study participants was isolated from peripheral blood, followed by PCR, gel electrophoresis and sequencing of the amplicons. Sequences were analyzed using MEGA X and genotyping was performed. Serum BDNF levels were assessed using ELISA. Results were analyzed using SPSS-21. Males were affected more as compared to female (PC substitution. Serum BDNF levels were significantly higher in cases as compared to controls (PC) was found with schizophrenia.
