Modulation of Apoptotic and Akt/Pi3k/Mtor Signaling Pathways in Glioblastoma Cells using Novel Compound 16(R and S)-Phenylamino-Cleroda3, 13(14) Z-Diene-15, 16 Olide (PGEA-AN)

Abstract:
Glioblastoma is the most lethal and frequently occurring type of central nervous system tumors. Inhibition of apoptosis and deregulated signaling pathways are the main culprits of disease. Drug resistance makes its prognosis hostile with median survival of 14-15 months. Therefore, finding new potential compounds against GBM is crucial. Terpenoids have influential biological activities including the anticancer. 16(R and S)-phenylamino-cleroda3, 13(14) Z-diene-15, 16olide (PGEA-AN) is a novel semi-synthetic derivative of naturally occurring diterpene which has shown apoptotic activity against neuroblastoma and should be explored for its therapeutic potential against GBM, which is also the aim of the study. MTT assay was performed to determine growth inhibitory effect of PGEA-AN and temozolomide (TMZ) against GBM U87 cells and IC50 was calculated. Phase-contrast microscopy and immunocytochemistry were done to investigate apoptotic morphology and Active caspase-3 and Bcl-2 expression. qRT-PCR was done to investigate the expression of proliferative markers. PGEA-AN and TMZ significantly inhibited growth of U87 cells (p0.05). The results of the study advocated that PGEA-AN holds growth inhibitory activity against GBM via induction of apoptosis and irrespective of Akt/PI3K/mTOR pathway.
