Abstract:
The purpose of this investigation is to understand the obesity-induced anxiety like behavior in rats. The study is designed to discuss the underlying mechanism of Salbutamol, a beta-adrenergic drug, in the management of obesity induced anxiety and protective role of Autumn Olive (Elaeagnus Umbellata. Obesity, the most occurring risk factor of metabolic syndrome, involves complex etiological links between the genetic, metabolic and neural frameworks. Antihistamines like Cetirizine and Fexofenadine play a critical role in weight gain. Beta2-adrenergic receptors combat obesity because they regulate the metabolism of adipose and skeletal muscle tissues. Male Albino Wistar rats (n=48) weighing 120±20gms were used in this investigation. Experimental rats were housed in controlled environment for adaptation. Rats were randomly assigned as Group 1 (n=24) fed with (NLC) and Group 2 (n=24) fed with high fat diet (HFD) for four weeks. Later, in next phase, the groups were sub-divided as; Control injected with Saline (0.9% NaCl) and tests injected with Cetirizine (1.0 mg/kg; CTZ) and Fexofenadine (3.0 mg/kg; FXF) respectively for 5 days. The route of administration for both vehicle and drug was intraperitoneal (i.p.). On the 6th day of experimentation, the control group was treated with Vehicle + Autumn olives and the other two groups prior treated with CTZ and FXF were then treated with salbutamol (1.0 mg/kg) + Autumn olives (100 mg/kg/ml) and Autumn olives ( 100mg/kg/ml) + Autumn olives (100 mg/kg/ml) respectively for 5 days. Physiological data in terms of Body weight, Food Intake and Growth rate) were monitored weekly. Behavioral responses were monitored using experimental models of Elevated Plus Maze and Light Dark Transition tests. Physiological assessments revealed significant decreases in the body weight, food intake and Growth rate of animals treated with SAL-AO. Elevated Plus Maze depicted significant increase in the numbers of entries in open arm of SAL-AO treated rats. Behavioral testing through light dark transition test depicted significant increases in the numbers of entries in the lit compartment of SAL-AO treated groups. Salbutamol, used as an anti-obesity drug, developed anxiety-like effects in rats. Supplementation of autumn olive primarily in high fat fed rats proven as an remarkable anti-obesity agent with no psychological adverse effects.
