Relationship between Genetic Variant of OXTR (rs53576) and MTNR1B (rs1387153) and Symptoms of Psychological Stress in Females with Gestational Diabetes Mellitus

Abstract:
The objective of the current study was to assess the association of the Oxytocin receptor (rs53576) and Melatonin Hormone receptor 1B (rs1387153) gene SNPs in women with GDM. Involvement of genetic mutations (Single nucleotide polymorphisms) in causation of stress and GDM is well established. Pregnancy is a stressful state, and Gestational Diabetes Mellitus (GDM) might make it adverse, however no such study is done in the local community in this context.This case control study`s goal was to identify the relationship of candidate genes (OXTR rs53576 & MNTR1B rs1387153) SNPs with psychological symptoms in women with GDM, it was conducted from May 1 to June 1 at the Department of Physiology, University of Karachi, in collaboration with the Department of Biological and Biomedical Sciences, Aga Khan University, and the Department of Obstetrics and Gynecology, Unit 8 Jinnah Post Graduate Medical Centre, Karachi, Pakistan. Genotypic analysis was done for all the study subjects (Macrogen, Korea). Perceived Stress Scale and Diabetes related Distress Scale were used to assess stress.: Using SPSS version 23 results were computed, and the mean sum of PSS score was 9.92±6.23, mean sum of DDS was 24.68±6.85, mean total DDS was 24.72±6.99, mean Emotional Burden score was 8.76±3.43, mean Physician Distress score was 5.68±1.86, mean Regimen Distress score was 6.82±2.29 and mean Interpersonal Distress score was 3.46±1.58, respectively among cases. A significant mean difference for Regimen Distress scores between AA and GG for OXTRrs53576 SNP was observed with p=0.04. Additionally, a significant mean difference in the sums of PSS (p=0.001) and DDS (p=0.001), total DDS (p=0.001), and emotional distress scores (p=0.001) between CC and TT genotypes for MTNR1B rs1387153 was noted. The results of this study show a trend of association between OXTR and MTNR1B genotype with stress and destress among gestational diabetes females.
