Use of real-time quantitative PCR in determining Phytophthora infestans (Mont) De Bary inoculum in soil.

Abstract:
Specific primers, based on ITS region of rDNA and SYBR Green, were used for on-line quantification of Phytophthora infestans in soil samples in a study conducted at Scottish Crop Research Institute, Invergowrie, Dundee (UK) in 2002. The starting copy numbers of the target sequences present in each reaction were calculated by comparing the Ct-values of unknown samples to the Ct-values of standards with known amount of DNA. The lowest detectable level of inoculum in 0.5 g soil samples was 30 oospores, which equals to 2pg/µl DNA. In a comparative study DNA was more efficiently recovered from clay (5.46pg/µl) than from sandy soil (0.84pg/µl). The results demonstrate that novel real time PCR is a powerful technique for evaluation of soilborne inoculum in potato fields.
