Antioxidative Role of Honey and L-Dopa against MPTP-induced Oxidative Stress in Adult Swiss Mice Model of Parkinson’s Disease

Abstract:
Oxidative stress is vital in the pathogenesis and progression of Parkinson’s disease (PD). 75% of dopaminergic (DA) neurons are already lost before the manifestation of the movement disorders. Moreover, one of the differential diagnoses of PD is its response to L-Dopa which does not prevent its progression. Hence, the need for neuroprotective therapy. Honey has been reported to be a radical scavenging gel and an enhancer of the antioxidant defence system. To investigate the antioxidative role of honey and L-Dopa against MPTP-induced oxidative stress.40 animals used for this study were divided into control and PD model groups of 20 animals each. Ten control animals received PBS, while the others received 1.5ml/kg body weight of honey and 80mg/ kg body weight of L-Dopa for 21 days. However, 10 of the PD model group were pretreated with HON and L-Dopa before PD induction. Behavioural studies were conducted two days after the induction, while the animals were euthanised seven days after. The result shows significantly (P=0.05) higher motor activities in the PD models pretreated with honey and L-Dopa. Furthermore, the pretreatment protected the midbrain against the gross neuronal damage and astrogliosis observed in the non-treated PD models. The expression of nuclear factor erythroid 2-related factor 2 (Nrf2) was also significantly upregulated (P=0.05) in the midbrain of the pretreated PD models. Pretreatment with honey and L-Dopa protected the DA neurons against oxidative stress via the mediation of the Nrf2 pathway.
