Abstract:
Application of Zinc fertilizers can make a significant contribution to achieve the goal of higher crop yields. Recently, nanoparticles have received considerable attention due to their increased uptake by plants as they are small in size and have higher rate of penetration through plant cell membrane. The present study investigated the effect of Zinc oxide nanoparticles (ZnONPs) on tomato plant (Lycopersicum esculentum L). For this purpose, Zinc nanoparticles were synthesized using water extract of Nigella sativa seeds and Zinc chloride solution. These were characterized by UV-Vis Spectrophotometer, Scanning electron microscope (SEM), Fourier Transform Infraerd Spectroscopy (FTIR). Petri plate experiment was performed to investigate the effect of ZnONPs on germination of tomato seeds. Tomato seeds were treated with various concentrations (i.e., 100, 300, 500 and 1000 mg/L) of ZnONPs. Seeds were planted in pots with contaminated soil containning chromium salt (potassium chromate) and treated with controls and Zinc nanoparticles treatments. Pot studies showed that the plants treated with 500mg/L concentration of ZnONPs showed maximum germination percentage, root length, shoot length and fruit number as compared to control plants. For molecular analysis, CDDP primers were selected from literature and were synthesized. DNA was extracted by cetytrimethyl amomoium bromide (CTAB) and quantification was done by nano drop method. After that, PCR was used to amplify DNA by specified CDDP (Conserved DNA-derived polymorphism) primers. The aim of present work was to study the behaviour of tomato samples treated with different salt stress conditions. CDDP analyses were employed to characterize the genetic relatedness among a commercial salt-sensitive tomato known for their tolerance to adverse conditions. Our results showed a general genotype dependent better growth and yield under all growth conditions. Furthermore, Principle Coordinate Analysis was done that differentiate tomato sample into two groups (Group 1 and Group 2). The highest polymorphism information content (PIC) values (0.96) were scored by CDDP-1, CDDP-2, CDDP-3, CDDP-4 and CDDP-5 respectively. The genetic stability among the tomato samples were studied as well.
