The hypothalamic neurosecretions of toad species Bufo melanosticus: chromatographic and biological characteristics.

Abstract:
Acetic acid-extracted peptides from 500 acetonedried hypothalami of toad, Bufo melanostricus were subjected for paper chromatography. The presence of 11 distinct ninhydrin positive bands was demonstrated. The eluted material in each band was oxidized with performic acid and hydrolysed with HC1 and reconstituted to obtain one and two dimensional chromotgrams for qualitative amino acid identification. The chromatographic fractions were assayed for oxytocic (with or without Mg++) and vasopressor activities. Bands 1-8 were characterized as peptides. Band 1-6 showed predominant rat pressor activity and no oxytocic activity whereas bands 7 and 8 exhibited both these activities. Bands 9 and 10 were found to depress blood pressure displayed oxytocin like activity both with or without Mg++ in the bath. Band 11 exhibited only rat oxytocic activity. Evidence is, therefore, presented for the presence of AVP and AVT only and MT seems to be absent in this species.
