Abstract:
Describes modified method wherein colour reagent is used without ferric chloride to eliminate non specific absorption. Six ml of glacial acetic acid was added to 0.1 ml of serum followed by 4 ml conc. sulphuric acid. The mixture was allowed to stand for 10 minutes and the density of colour was measured against a water blank. This density was subtracted from the colour obtained by ferric chloride method to correct the non-specific absorption. As the non-specific absorption is due to serum constituents no correction is applied to this cholesterol standard.
