DNA extraction from plant sources and study of its inter-actions with metal complexes/salts.

Abstract:
A quick, simple and reliable method has been developed for extraction of DNA from carambola, olive, tomato, potato, onion, brinjal, and clean DNA from carambola, onion and karela, respectively. Binding of the complexes (+)[Co(en)3]I3·H2O, (-)[Co(en)3]I3·H2O, (+)[Co(en)3][(+)- tart]Cl·5H2O, Na[Cr(acac)2)(ox)]·2H2O, [Cu(NH3) 4(H2O) 2](NO3)2, [Cu(en) 2(H2O) 2](NO3)2 and [Fe(phen)3] and also of the metal salts CuCl2·2H2O, and FeCl2·4H2O with DNA extracted from onion or brinjal have been investigated by spectroscopic as well as viscometric methods under identical experi-mental conditions. Possible mechanism of interactions of these complexes with DNA have been dis-cussed. The results indicate that [Cu(NH3)4(H2O)2](NO3)2, [Cu(en)2(H2O)2](NO3)2, [Fe(phen)3] com-plexes bind to DNA more effectively than (+)[Co(en)3]I3·H2O, (-)[Co(en)3]I3·H2O, (+)[Co(en)3][(+)- tart]Cl·5H2O and Na [Cr(acac)2)(ox)]·2H2O complexes, respectively. The copper and iron complexes are found to be keen binders of DNA with the binding constants in the order of 105. Analysis of the results suggest that binding ability of these complexes or of salts varies in the order [Cu(NH3)4(H2O)2](NO3)2> [Fe(phen)3]> [Cu(en)2(H2O)2](NO3)2> FeCl2·4H2O> (-)[Co(en)3]I3·H2O> (+)[Co(en)3]I3·H2O> (+)[Co(en)3][(+)-tart]Cl·5H2O> Na[Cr(acac)2)(ox)]·2H2O > CuCl2·2H2O.
