Transformation of forage legume Onobrychis vicifolia using binary vector in Agrobacterium rhizogenes.

Abstract:
A binary vector of A. rhizogenes was constructed, A4T (p81121), by mobilizing the plasmid pB1121, harbouring beta-glucuronidase gene (GUS) and neomycin phosphortransferase II (NPTII) gene into A. rhizogenes strain A4T. The binary vector A4T (pB1121) was used to transform hypocotyls of 0. viciifolia. Transformed roots were screened for their ability to grow on kanamycin containing medium and for GUS activity. The latter was detected using fluorometric and histochemical assays, and by DNA-DNA hybridization. Plants of 0. viciifolia were regenerated from these transformed roots. Histochemical assay showed beta-glucuronidase activity in petioles and leaf sections. Transformation efficiency was compared with two other A. rhizogenes strains A4T and A4TIII.
