Seed priming improves germination and seedling growth of oat (Avena sativa L). Under salt stress environment

Abstract:
Oat is an important winter fodder crop grown all over the world. Poor oat seed germination is a problem in salt-affected soils. Seed priming is a technique that can improve germination and promote early seedling establishment by overcoming salt stress. It helps in imparting the initial vigor to the seedlings which is essential for better seedling performance. The experiment was conducted in Agronomy Lab., College of Agriculture, University of Sargodha, using CRD factorial having seven priming treatment and five salt stress levels and replicated thrice. The seeds were soaked in solution of salicylic acid0.4mML-1, ascorbic acid 1.50mML-1, gibberellic acid 0.3mML-1, polyethylene glycol 15%, potassium 1%, calcium chloride 1% and in distilled water for 12hours, no priming was kept as control. After that, seeds were planted on petri dishes, and salt stress was given using NaCl solutions of 40, 60, 80, and 100mM-1. The data regarding seed emergence and seedling growth was recorded according to standard protocol and statistically examined using Fisher's analysis of variance method, Turkey’s honestly significant difference test (HSD) was used to compare treatment means at 5% probability level. Data showed the significant effects of priming and salt stress treatments on seed germination and seedling characteristics of oat. Highest germination (82.83%), speed of germination(10.52), mean daily germination (3.16%), lowest time to start germination, mean germination time (0.81%) and highest shoot length (4.78cm), seedling root length (5.14cm), seedling vigor index (825.68), seedling fresh shoot weight(31.58mg), seedling fresh root weight (28.43mg), dry shoot weight (13.97mg),dry root weight (12.69mg) and seedling roots number (13.27) were recorded when seed were primed with salicylic acid only without salt stress but priming of salicylic acid improves tolerance under salt stress of 40 mM-1 and all germination and seedling parameters were also improved.
