Genotype analysis of beta-casein gene variants (A1/A2) in Sahiwal cattle breeds using hair follicles

Abstract:
Cow’s milk has been proved to be rich in many nutritional elements especially proteins and thus makes it important part of human diet. Casein is most abundant with 80% portion of bovine milk proteins. Out of 12 variants of ß-casein, abundance of A2 ß-casein gene variant in milk makes it beneficial for health as A1 ß-casein gene variant is linked with many health problems like lactose intolerance. Single nucleotide polymorphism at exon 7 of Bos-taurus chromosome 6 (BTA6) results in the substitution of Proline in A2 to Histdine in A1. A2 variant of ß-casein is common in Sahiwal cattle but A1 variant also occur in heterozygous form. For the genotype analysis of beta-casein variants A1/A2 in Sahiwal cattle of farms around Lahore were analyzed. Hair samples were collected but due to various factors like DNA degradation and cross contamination during experimental process, no satisfactory DNA extraction results from hair follicles could be obtained. DNA extraction from blood was done by organic method. DNA quantification was estimated by Nanodrop photo-spectrometry. PCR-RFLP was performed with reported restriction primers (CASB67L and CASB122R) were used along with restriction buffer in a volume of 30ul. Bands on 2.5% Agarose gel confirmed successful restriction. SNP analysis of ßCSN2 gene’s exon 7 region was last step for genotype analysis. Sequencing results were interpreted using BLASTn and Chromas software. Results showed that A2 beta-casein variant is common in Sahiwal breed but A1 variant in low frequency also occur in heterozygous form. Inbreeding programs of cattle breeds based on A1/2 beta-casein variant should be measured.
