A rapid Protocol for Extraction of Podophyllotoxin (An Anticancerous Compound) from Podophyllum hexandrum Royle.

Abstract:
Microwave assisted extraction is a modern and environment friendly approach for rapid and efficient extraction of secondarymetabolites of primaryimportance from plants. Podophyllum hexandrum is an important medicinal plant. Aryltetralin compound is obtained from this plant which is used as a precursor of a number of anticancer drugs on commercial scale. In present investigation one of the conventional extraction methods (cold maceration) and one of the modern method (microwave assisted extraction [MAE]) were compared for extraction of podophyllotoxin. Solvents used for this purpose were ethanol, ethyl acetate and n-hexane. MAE gave maximum amount of 190 mg and cold maceration gave 35.53 mg per gram of plant material with ethanol. Both the extraction methods were evaluated by quantitative analysis of podophyllotoxin using HPLC. The quantity was estimated to be higher with MAE (76 %) at 400W for 180 seconds as compared to cold maceration (67%) after 10 hours. So, Microwave assisted extraction proved to be a better analytical methods as it takes lesser time and release less energy and hazardous chemicals. It can be recommended for commercial production of podophyllotoxin as it is environment friendly.
