Optimization of auxins and cytokinins to induce callus in polianthes tuberosa

Abstract:
The aim of study is to optimize conditions for callus induction of P. tuberosa employing rhizome as an explant. Various concentration of 2,4-D, kinetin and BAP alone or in combination were experimented to induce callus under aseptic controlled conditions on MS medium. The maximum callus induction (100%) was obtained on MS medium containing 0.05 mg/L kinetin and combination of 2,4-D and BAP (0.05+ 0.05 mg/L and) after 40 days of inoculation. Minimum callus was induced in 2,4-D (0.04, 0.05). The minimum callus was appeared in the Kinetine (0.02, 0.03 mg/L), 2,4-D and BAP (0.01+ 0.06 and 0.01+ 0.06 mg/L). The color and texture of callus were also recorded. The study can be useful for in vitro propagation of P. tuberose to meet the demand for ornamental, medicinal and industrial purpose.
