Biological activities of Vincetoxicum stocksii (L.) the endemic plant of district Zhob, Balochistan, Pakistan

Abstract:
People of the many countries of the world are using medicinal plants for their ailments since pre historic times. During this investigation we evaluated the traditional claims related to Vincetoxicum stocksii. The plant was collected from Baluchistan, extracted and fractionated to segregate the components. Further, UHPLC, and FTIR analysis was performed. Regarding quantitative analysis, flavonoids, terpenes and saponins were detected. The plants fractions were further processed for in vitro evaluation. Initially during antioxidant assays, With the DPPH assay a significant activity was seen in ethyl acetate part with (IC50 0.29 mg/mL), preceeded by aqueous (IC50 0.431 mg/mL) and methanolic parts (IC50 0.69 mg/mL).During the FRAP assay for V. stocksii,, the methanolic part showed FRAP value (740.2 ± 0.12mg/gGAE), lagged by ethyl acetate part (468.7 ± 0.01mg/gGAE). During antimicrobial investigations, the E. coli showedvividly superior activty in methanolic (MIC/MBC=0.019/0.078 mg/ml) and ethyl acetate parts (MIC/MBC = 0.078/0.156 mg/mL). Likewise, in case of Pseudomonas aeroginosa, the n-hexane fraction showed significant activity (MIC/MBC =0.625/ 1.25 mg/mL) followed by (MIC/MBC = 0.625/ 0.625 mg/mL) whereas all other fractions were not active in the tested concentration. In case of antidiabetic assays overall, all tested fractions were active, however the methanolic (IC50 0.28mg/mL) and ethyl acetate fractions (IC50 0.28mg/mL) were more active against alpha-glucosidase. Finally, in case of 15-lox inhibition, mild to moderate activity have been observed in methanolic fraction (46%) subsequently by ethyl acetate part (42%). All other fractions were inactive under testing concentrations. It was concluded that V. stocksii posesses antioxidant, antimicrobial, antidiabetic and antioxidant activites.
