Abstract
Bacillus subtilis being one of the most widely utilized species due to its capacity to produce various enzymes on a commercial scale. Among these, a-amylase (EC 3.2.1.1) holds particular importance for applications in the textile, detergent, food and beverage industries. In this study, a-amylase-producing Bacillus subtilis was isolated from rice field soil, and its enzyme characteristics and kinetics were systematically investigated under different physical and chemical conditions. Using starch as the main carbon source and peptone and yeast as nitrogen sources increased the synthesis of alpha amylase synthesis. The medium was incubated for different fermentation time (24 hours to 96 hours at a different temperature between 30°C and 60°C with an initial pH of 7.0. The enzyme showed its optimal activity after 24h hour of fermentation time at 40°C at optimum pH (7.0). These findings suggest that the a-amylase from Bacillus subtilis possesses favorable characteristics such as stability of enzyme, rapid catalytic reaction and enhanced activity under optimized fermentation conditions. Such properties highlight its potential for diverse industrial applications, particularly in processes requiring efficient starch hydrolysis.
